Psychoneuroendocrinology:犬尿氨酸代谢与炎症引起的抑郁情绪

炎症对情绪和行为有重要的生理影响。据推测,犬尿氨酸代谢是连接炎症和抑郁情绪的途径,部分是通过犬尿氨酸代谢物对中枢神经系统中谷氨酸神经递质的影响。本研究评估了在炎症诱导的抑郁情绪的人类模型中,在炎症挑战

炎症对情绪和行为有重要的生理影响。据推测,犬尿氨酸代谢是连接炎症和抑郁情绪的途径,部分是通过犬尿氨酸代谢物对中枢神经系统中谷氨酸神经递质的影响。本研究评估了在炎症诱导的抑郁情绪的人类模型中,在炎症挑战(内毒素给药)的反应下,犬尿氨酸和相关化合物的循环浓度是否发生急性变化,以及这种代谢物的变化是否与情绪变化有关。

研究纳入115名成年人,随机接受内毒素或安慰剂。在干预前、干预后2小时和6小时反复测量情绪(情绪状态简介)、血浆细胞因子(白细胞介素-6、肿瘤坏死因子-α)和代谢物(炔诺酮、色氨酸、炔诺酮酸、喹啉酸)浓度。使用线性混合模型评价情绪、犬尿氨酸及相关化合物、细胞因子之间的关系。

 

结果显示,与安慰剂相比,犬尿氨酸、犬尿酸和色氨酸(但不是喹啉酸)浓度对内毒素的反应发生急性变化(p's均<0.001)。犬尿氨酸、犬尿酸和色氨酸浓度在基线时均与细胞因子浓度无关,但随着时间的流逝,在对内毒素的反应,三者均与细胞因子浓度显著相关。喹啉酸浓度与细胞因子浓度在内毒素治疗前或治疗后均无关。在接受内毒素的人中,犬尿氨酸(p = 0.049)和喹啉酸(p = 0.03)与抑郁情绪呈正相关,尽管这些发现不能经受多重测试的校正。

 

综上所述,该研究结果表明,色氨酸和犬尿氨酸途径代谢物的变化并没有介导细胞因子和抑郁情绪之间的关系。有必要进一步的工作来明确人类从炎症导致抑郁情绪的途径。
