J Endod：合成黏土类模拟低氧水凝胶在牙髓再生中的应用：体外对牙髓来源细胞活性的影响及释放动力学

摇溶合成黏土已经成功的应用于再生医学的组织工程中。然而，这种黏土对于牙髓的影响，特别是这种材料通过低氧模拟因子(HMAs)与低氧相结合后的影响还未可知。这篇研究的目的是为了揭示合成黏土类水凝胶对牙髓来源细胞(DPCs)的影响以及评估它们HMAs的释放情况。研究使用刃天青类的毒性测定、活-死染色和溴化3-（4，5-二甲基噻唑-2-基）-2，5-二苯基四唑染色，将人DPCs接种于5%-0.15%的合成黏土类水凝胶上，也接种于负载了HMAs二甲基肟基甘氨酸（DMOG），去铁胺，L-含羞草碱和CoCl2的水凝胶上，进行细胞活性的评估。另外，制取负载HMAs水凝胶的上清液。检测施加上清液刺激的DPCs的血管内皮生长因子(VEGF)水平以揭示细胞对HMAs的反应。结果显示，合成黏土类水凝胶并不影响DPCs的活性。水凝胶上可观察到单层细胞及细胞团簇的形成。对培养于负载HMAs水凝胶上的DPCs施加上清液刺激时，未检测到VEGF水平的显著上调。负载DMOG水凝胶的上清液刺激DPCs 1小时可明显诱导VEGF表达的上调，而去铁胺，L-含羞草碱和CoCl2的作用并未达到显著性水平。结论：合成黏土类水凝胶代
