CXCL14特发性肺纤维化Hh信号标志物

由此可见，CXCL14是一个全身性的生物标志物，可以用来识别Hh通路活性增加的IPF患者，并且监控IPF患者进行Hh拮抗剂治疗的药效学作用。

特发性肺纤维化（IPF）与发育信号通路中分子的异常表达有关，这些分子包括Hedgehog（Hh）信号（通路）。由于Hh信号通路有助于多种促纤维化过程的发生发展，抑制Hh信号通路可能是IPF的一种治疗选择。然而，并没有非侵入性的生物标志物可用于监测肺脏Hh信号通路的活性。

近日，胸部疾病领域权威杂志Thorax上发表了一篇研究文章，研究人员检测了IPF患者和对照者肺脏活检标本、小鼠肺脏、离体培养的接受Sonic hedgehog（SHh）处理的成纤维细胞、IPF患者和对照者血浆以及接受Hh抑制剂vismodegib治疗的肿瘤患者治疗前后血浆中蛋白表达情况。

研究人员从IPF患者采集的肺组织中发现Hh相关基因的表达程度相比于对照者显著增加。而在IPF患者和离体培养的接受SHh处理的成纤维细胞中表达上调最显著的是CXCL14，该基因编码可溶性分泌趋化因子，其表达的可被体外vismodegib处理所抑制。通过小鼠肺脏过表达SHh可以诱导CXCL14的表达。IPF患者循环中CXCL14蛋白水平明显高于对照者。在癌症患者中，接受vismodegib治疗后循环CXCL14的水平明显降低。

由此可见，CXCL14是一个全身性的生物标志物，可以用来识别Hh通路活性增加的IPF患者，并且监控IPF患者进行Hh拮抗剂治疗的药效学作用。
