Sci Rep:麦角甾醇过氧化物诱导人肺腺癌细胞的ROS依赖性凋亡和自噬

EP通过A549细胞中的线粒体损伤诱导半胱天冬酶依赖性细胞凋亡。

作为正在进行的寻找来自微生物新型治疗结构的一部分,对海洋真菌Phoma sp.的化学分析分离产生了麦角甾醇、麦角甾醇过氧化物(EP)和9、11-脱氢甾醇过氧化物(DEP)。

生物测定结果表明,三种分离物降低各种癌细胞的活力,其中EP在人肺癌细胞系A549细胞中活性最高。EP通过A549细胞中的线粒体损伤诱导半胱天冬酶依赖性细胞凋亡。此外,ROS生成酶抑制剂和抗氧化剂N-乙酰半胱氨酸减弱了EP诱导的ROS生成和凋亡,这表明ROS在EP介导的A549细胞凋亡中起重要作用。此外,还观察到EP诱导的ROS依赖性自噬,其减弱了A549细胞的凋亡。另一方面,EP通过减弱NLRP3炎性体活性降低了LPS/ATP诱导的A549细胞增殖和迁移。此外,EP显示与抗肿瘤药物索拉非尼在A549细胞活性抑制上有协同细胞毒作用。此外,蛋白质微阵列和蛋白质印迹分析显示EGFR、HSP27、MEK5、AKT1、mTOR、Smad2、Smad3、TAB1、NF-κB和HIF1-α的蛋白水平下降,而p-p38α、p-ERK1/2、p-JNK、纤连蛋白和p27水平升高。

总的来说,这项研究结果表明,EP有助于开发肺癌并发症的候选疗法。
