炎症发生和血栓形成会加剧局部缺血事件发生后导致的脑组织损伤。细胞外ATP和ADP水平升高与细胞损伤、炎症发生和血栓形成有关。在白细胞和内皮细胞膜上表达的核苷三磷酸二磷酸水解酶-1(即CD39),通过磷酸水解ATP/ADP抑制血小板活化和白细胞浸润。

近期美国学者发表研究文章于著名血管杂志Circulation,以研究CD39增加在在体脑缺血模型中的作用。研究构建了表达人CD39(hCD39)的转基因小鼠。

与野生型小鼠相比,全身hCD39转基因小鼠(n = 9)和髓系CD39-过表达小鼠(n = 6)脑梗死体积明显减小。使用流式细胞术分析缺血侧和对侧脑半球的白细胞。虽然对侧脑半球具有相同数量的巨噬细胞和嗜中性粒细胞,但全身hCD39转基因小鼠的缺血侧脑半球浸润较少(n = 4)。与野生型小鼠相比,转基因小鼠有较少的神经功能损伤(n = 6)。

本研究首次报道在小鼠体内CD39过表达可保护机体防止卒中,减少白细胞浸润、梗死体积以及神经功能缺损。全身或髓系细胞中CD39过表达抑制缺血后白细胞浸润,减少卒中诱发的神经损伤。
