牙周炎是一种慢性炎症性疾病，是导致成人牙齿脱落的主要原因。关于桑树提取物（MSE）在牙周病中的抗炎活性的研究十分有限。本研究旨在探讨MSE对牙龈卟啉单胞菌脂多糖（LPS）刺激的人牙周膜成纤维细胞（hPDL）中牙周致病菌的生长和白细胞介素（IL）-6和IL-8表达的抑制作用。

通过盘扩散、最小抑制浓度和最小杀菌浓度方法测试MSE对牙龈卟啉单胞菌和伴放线聚集杆菌的抗菌活性。通过MTS测定法测定牙龈卟啉单胞菌LPS和MSE对hPDL成纤维细胞的细胞毒性。使用逆转录-qPCR和酶联免疫吸附测定分别测量hPDL成纤维细胞中细胞因子（IL-6和IL-8）mRNA和蛋白的表达。

结果显示，MSE对牙龈卟啉单胞菌和伴放线聚集杆菌都具有抗菌活性，抑菌圈分别为10.00±0.33mm和17.33±0.58mm。MSE对牙龈卟啉单胞菌的MIC和MBC值为62.5μg/ml。MSE对放线菌伴放线菌的MIC和MBC值分别为250μg/mL和500μg/ml。牙龈卟啉单胞菌LPS显示介导hPDL成纤维细胞中促炎细胞因子的表达。然而，浓度为2.5和5.0μg/ml的MSE可显著抑制牙龈卟啉单胞菌LPS诱导的IL-6和IL-8 mRNA和蛋白质表达（p <0.05）。

本研究表明，MSE对牙龈卟啉单胞菌和伴放线聚集杆菌都具有抗菌活性。MSE抑制牙龈卟啉单胞菌LPS刺激的hPDL成纤维细胞中IL-6和IL-8的表达，表明具有抗炎作用。因此，MSE或是治疗牙周炎的潜在辅助剂。
