鳞状细胞肺癌约占非小细胞肺癌的25%，此类患者预后较差，可选择的治疗方式有限。FGFR1过表达是鳞状细胞肺癌最常见的致癌原因之一，约在20%的患者中存在。AZD4547是选择性FGFR1-3抑制剂，在鳞状细胞肺癌细胞系及异种移植模型中具有抗肿瘤活性。CLIN CANCER RES近期发表了一篇文章， 报道了AZD4547治疗鳞状细胞肺癌患者的临床试验结果。

鳞状细胞肺癌约占非小细胞肺癌的25%，此类患者预后较差，可选择的治疗方式有限。FGFR1过表达是鳞状细胞肺癌最常见的致癌原因之一，约在20%的患者中存在。AZD4547是选择性FGFR1-3抑制剂，在鳞状细胞肺癌细胞系及异种移植模型中具有抗肿瘤活性。CLIN CANCER RES近期发表了一篇文章， 报道了AZD4547治疗鳞状细胞肺癌患者的临床试验结果。研究纳入的患者为接受过治疗的Ⅳ期FGFR1过表达鳞状细胞肺癌患者。使用FISH法鉴定FGFR1过表达（FGFR1：CEP8≥2）。主要的研究终点为安全性/耐受性。次要的研究终点包括抗肿瘤活性，药代动力学，药效学和分子分析。研究共纳入15例FGFR1过表达患者。最常见的治疗相关不良反应为胃肠道和皮肤反应。3例患者出现3级及以上不良反应。13例患者可以进行影像学治疗反应评估。总反应率为8%。15例患者中2例患者在12周出现无病情进展，平均总生存期为4.9周。分子检测，包括二代测序，基因表达分析和FGFR1免疫组化分析表明基因扩增与表达，潜在的疗效基因修饰和8p11扩增异质性关系不明显。文章最后认为，AZD4547在剂量为80mg口服每天2

Cancer Discov.：PI3K畸变致使鳞状细胞肺癌向脑转移
