加州大学洛杉矶分校的研究人员Cun-Yu Wang博士带领的研究小组将开发一种新的治疗药物,不仅会减慢骨质破坏,而且还会抑制骨髓炎症,促进骨形成。

研究人员发现,在骨质疏松症和骨老化小鼠模型中,一种生长因子Wnt4会被分泌到骨髓,能在一定程度上防止骨损失,其效应是通过抑制NF-κB信号传导来实现的。

研究小组此前发现,Wnt4基因蛋白可促进骨形成,在新的研究中,他们构建了成骨细胞或骨形成细胞Wnt4基因过表达的转基因小鼠。正如所预期的那样,与对照组相比,这些小鼠表现出骨质量改善,表现出较少的骨损失。出人意料的是,研究小组发现,Wnt4蛋白不仅促进成骨细胞活性,同时也可以抑制破骨细胞或骨吸收细胞的活性,以及骨髓中的炎症。

为了理解Wnt4蛋白的作用机制,研究人员分析其对NF-κB信号传导的影响,后者在破骨细胞形成和骨髓炎症中发挥关键作用。研究发现NF-κB信号过度活跃的小鼠出现关节炎症状,表现出骨质流失,但这些小鼠与Wnt4基因过表达的转基因小鼠杂交后,上述症状得到有效缓解。Wnt4转基因小鼠也显示年龄相关骨质量下降速度较慢。该团队接着证实了Wnt4蛋白在体外能抑制破骨细胞形成。

最后,为了研究Wnt4对骨质疏松症的治疗用途,该团队利用骨质疏松症小鼠,将重组Wnt4蛋白注射到这些小鼠。他们发现Wnt4蛋白对小鼠骨质疏松有积极的治疗效果,骨质流失显著减少。这些研究提示了重组Wnt4蛋白可能是治疗骨质疏松症与衰老相关骨质流失的有效药物。
