胞内细菌,病毒等细胞内感染病原体能够利用多种策略适应以及改变宿主细胞的胞内环境,逃脱宿主的免疫防御。许多胞内病原体利用细胞生理性的内吞囊泡途径侵染细胞,并适应了细胞内吞囊泡的酸化环境以达到其感染的目的。外源性的化学物质如氯化铵和氯喹等可以通过改变细胞内吞囊泡的酸化环境而抑制某些病毒和胞内寄生虫的感染,但宿主细胞是否存在相应的防御策略,内源性效应分子机器的身份以及调控网络一直是人们努力探究的重要问题

中科院昆明动物研究所生物毒素与人类疾病课题组在研究员张云带领下,前期从两栖动物大蹼铃蟾(Bombina maxima)中发现了脊椎动物第一个新型孔道形成蛋白和三叶因子复合物betagamma-CAT (发明专利ZL200810058028.5),并发现该内源性蛋白质机器具有刺激细胞囊泡化形成的特性,在细胞内吞/溶酶体途径中形成膜通道并激活炎症小体(Proc Natl Acad Sci USA 2014,111,6702,封面故事)。该课题组李盛安和刘龙等进一步深入揭示该复合物具有调控细胞内囊泡酸化环境的功能。在宿主免疫防御中,以细胞内病原体单核增生李斯特菌(Listera monocytogenes)为模型的研究表明该蛋白质机器可以限制李斯特菌从胞内囊泡的逃逸,并同时激发细胞外排驱逐胞内感染的病原体,有效抵御致死性的李斯特菌感染。论文以 Host pore-forming protein complex neutralizes the acidification of endocytic organelles to counteract intracellular pathogens为题在线发表在美国传染病学会会刊 Journal of Infectious Diseases上 。副研究员李盛安和博士研究生刘龙是文章第一作者,张云是文章通讯作者。

该研究不仅揭示了脊椎动物防御和清除细胞内病原体的新策略和效应分子,也为深入解析细胞囊泡化形成和调控的基础细胞生物学问题以及相关的生理病理作用提供了新思路和新线索,对于研发新的疾病治疗策略和药物也具有实际应用的意义。研究获得国家基金委-云南联合基金重点项目以及“云岭学者”的资助。
