将AmeloD定位于小鼠染色体1q32上。系统发育分析表明，AmeloD属于achaete-scute complex-like（ASCL）基因家族，是ASCL5的同源基因。 AmeloD在内部釉质上皮细胞（IEE）中独特表达，但其表达在IEE细胞分化为成釉细胞后被抑制。此外，AmeloD表达显示出与牙齿上皮中的上皮细胞特异性细胞-细胞粘附分子E-钙粘蛋白的反向表达模式。AmeloD在牙齿上皮细胞系CLDE细胞中的过度表达导致E-钙粘蛋白抑制。我们发现AmeloD与E-钙粘蛋白基因的近端启动子区域中的E-box顺式调节元件结合。这些结果揭示AmeloD在IEE细胞中作为E-钙粘蛋白转录的抑制剂起作用。

总之，我们的研究表明，AmeloD是一种新型牙齿特异性bHLH转录因子，可通过抑制IEE细胞中的E-钙粘蛋白来调节牙齿发育。
