作者对27例接受切除术的DGA患者的肿瘤样本进行了全外显子组测序。在DGA细胞系和患者肿瘤中分析KMT2C情况。研究结果表明,KMT2C是最常见的突变基因。通过免疫组化检测135例接受胃切除的DGA患者的肿瘤KMT2C表达情况,KMT2C表达与肿瘤分期较晚和总体生存率较差呈负相关。在HFE-145胃上皮细胞中通过KMT2C shRNA敲低KMT2C促进上皮-间质转化(EMT)。KMT2C敲低后胃上皮细胞的迁移和侵袭能力增加47-88倍。在无KMT2C表达的DGA细胞系MKN-45和SNU-668中,重新表达KMT2C可以降低EMT相关蛋白的表达,使细胞迁移能力减少52%至60%,并使细胞侵袭能力减少50%至74 %。EMT可导致肿瘤干细胞(CSC)表型增加。DGA细胞系中重新表达KMT2C可以通过促进DNA损伤和凋亡逆转CSC化疗耐药。

文章最后认为,KMT2C在特定DGA人群中经常发生突变。DGA中的KMT2C缺失促进EMT并且与总生存较差有关。
