Science:生长调节蛋白mTORC1对接在溶酶体表面的结构基础被解析

最近,研究人员通过冷冻电子显微镜确定了具有Rag-Ragulator的Raptor超级复合体的结构,分辨率为3.2Å。

已有的研究显示,mTORC1蛋白激酶复合物通过响应营养物和生长因子来调节生长。 营养物质促进其向溶酶体表面的转运。在溶酶体表面,其Raptor亚基与Rag GTPase-Ragulator复合物相互作用。

营养物会使得异二聚体Rag GTPases在四种不同的核苷酸结合状态之间切换,而只有在一种状态下(RagA / B•GTP–RagC / D•GDP),mTORC1能够在溶酶体上缔合。

最近,研究人员通过冷冻电子显微镜确定了具有Rag-RagulatorRaptor超级复合体的结构,分辨率为3.2Å Raptor α-螺线肌醇直接检测RagA的核苷酸状态,而Raptor爪子穿过GTPase结构域,以检测RagC的核苷酸状态。

能够破坏Rag-Raptor结合的突变会抑制mTORC1的溶酶体定位和信号传导。

通过与其激活物Rheb结合的mTORC1的结构进行比较,研究人员开发了定位在溶酶体上的活性mTORC1的模型。
