Mol Med Rep:SNHG12上调可促进肝细胞增殖和肝再生

在部分肝切除术(PH)之后,开始了肝再生的复杂过程,其中包括同步诱导肝细胞增殖。肝细胞增殖可以通过多种刺激来调节,包括长的非编码RNA(lncRNA)和Wnt /β-catenin信号传导,尽管lncRNA / Wnt在肝脏再生中的潜在机制尚不清楚。在本研究中,我们鉴定了与肝脏再生相关的功能性lncRNA,并在体内和体外描述了其功能。

 

2/3 PH后lncRNA小核仁RNA宿主基因12(SNHG12)的表达在各个时间点上调。在不同浓度的肝细胞生长因子(HGF)处理的正常肝细胞系中,SNHG12的表达也增加了。从功能上讲,SNHG12在体外和体内均能增强肝细胞的增殖,并且过表达SNHG12的小鼠的肝脏/体重比显著高于对照小鼠。SNHG12的过表达促进了肝细胞中Wnt /β-catenin信号的激活。此外,对Wnt /β-catenin信号的特异性抑制明显减弱了SNHG12诱导的肝细胞增殖和受影响的肝脏/体重比。

 

总体而言,本研究的结果表明SNHG12通过激活Wnt /β-catenin信号传导促进肝脏再生。因此,调节SNHG12/Wnt轴的药物可能对PH后的肝脏再生有益。
